Biotin-dPEG-N3

Biotin-dPEG-Azide (Biotin-dPEG-N3) includes discrete polyethylene glycol (dPEG) linkers containing a biotin group and a terminal azide (N₃) functionality. These reagents combine the strong biotin–avidin/streptavidin interaction with bioorthogonal azide click chemistry, providing versatile tools for bioconjugation, affinity labeling, protein modification, biomolecule immobilization, and molecular probe development.

The terminal azide reacts with terminal alkynes through copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) or with strained cyclooctynes such as DBCO and BCN through copper-free strain-promoted azide-alkyne cycloaddition (SPAAC).

The biotin functionality enables high-affinity interaction with avidin, streptavidin, and neutravidin, allowing biotinylated conjugates to be captured, detected, purified, or immobilized.

Unlike conventional polydisperse PEGs, dPEG linkers have defined PEG chain lengths and molecular structures, providing precise control over spacer length between the biotin and azide functionalities.
