General Description
10-Hydroxycamptothecin (HCPT) is a semi-natural camptothecin analog that exhibits potent antitumor activity through selective inhibition of DNA topoisomerase I. By stabilizing the topoisomerase I-DNA cleavage complex, HCPT prevents DNA religation, resulting in DNA damage, inhibition of DNA replication, and apoptosis of rapidly proliferating tumor cells.
Because of its limited water solubility and lactone ring instability under physiological conditions, HCPT has been extensively investigated in liposomes, polymeric micelles, PEGylated nanoparticles, lipid nanoparticles (LNPs), hydrogels, and other nanocarrier systems to improve its stability, bioavailability, and tumor-targeting capability.
