General Description
DOPE is one of the most commonly used helper lipids in lipid-based drug delivery systems. The lipid contains two oleic acid chains attached to a glycerol backbone and a phosphoethanolamine head group. Unlike phosphatidylcholines, DOPE tends to adopt non-bilayer hexagonal (HII) phases, which facilitate membrane destabilization and fusion.
This unique property makes DOPE particularly valuable in gene delivery, mRNA delivery, siRNA delivery, liposome formulation, and lipid nanoparticle development. DOPE is frequently combined with cationic lipids, ionizable lipids, cholesterol, and PEG lipids to improve intracellular delivery efficiency.
