General Description
Maleimide-PEG-PLGA-Amine (MAL-PEG-PLGA-NH₂) is a dual-functional biodegradable polymer incorporating polyethylene glycol (PEG), poly(lactic-co-glycolic acid) (PLGA), a maleimide (MAL) functionality, and a primary amine (NH₂) functionality.
The PLGA component provides a biodegradable hydrophobic polymer domain suitable for nanoparticle formation and drug encapsulation, while PEG provides hydrophilicity, steric stabilization, and a flexible spacer for surface presentation of functional groups.
The maleimide group reacts preferentially with thiols (–SH), including cysteine-containing peptides and proteins, to form stable thioether linkages. This enables conjugation of targeting peptides, antibodies, proteins, and other thiolated molecules.
The amine functionality provides a second chemically addressable group that can react with NHS esters, activated carboxylic acids, aldehydes, fluorescent dyes, drugs, and other amine-reactive compounds.
The presence of two different reactive functionalities makes MAL-PEG-PLGA-NH₂ particularly useful for orthogonal and multistep conjugation strategies, allowing different molecules to be introduced onto the same polymeric platform.
