General Description
PDLLA-PEG-PDLLA is an amphiphilic ABA triblock copolymer composed of poly(D,L-lactide) (PDLLA) and polyethylene glycol (PEG). The amorphous PDLLA segments provide a biodegradable hydrophobic matrix suitable for encapsulating hydrophobic therapeutics, while the PEG middle block enhances aqueous solubility, colloidal stability, and biocompatibility.
Unlike PLLA-PEG-PLLA, the PDLLA segments are amorphous because they are derived from racemic D,L-lactide. This results in reduced crystallinity, faster hydrolytic degradation, and improved flexibility, making PDLLA-PEG-PDLLA particularly attractive for controlled drug release, injectable formulations, thermosensitive hydrogels, and tissue engineering applications.
PDLLA-PEG-PDLLA readily self-assembles into polymeric micelles and nanoparticles in aqueous media and is frequently used as a building block for biodegradable nanocarriers and responsive biomaterials. Polymer composition, molecular weight, and block ratio can be tailored to optimize degradation rate, drug loading capacity, and release kinetics.
