General Description

PDLLA-PEG-Folate (PDLLA-PEG-FA) is a functional biodegradable block copolymer consisting of poly(D,L-lactide) (PDLLA), a hydrophilic polyethylene glycol (PEG) spacer, and a terminal folate (folic acid) targeting group.

The hydrophobic PDLLA block provides an amorphous, biodegradable polymer domain suitable for encapsulating hydrophobic compounds and forming polymeric nanoparticles, micelles, and other self-assembled nanostructures. The PEG segment improves aqueous dispersibility and colloidal stability while presenting the folate ligand at the surface of the resulting nanocarrier.

Folate is widely used as a targeting ligand because folate receptors are overexpressed on certain cancer cells and other cell types. Incorporation of PDLLA-PEG-Folate into polymeric nanoparticles can provide folate receptor-targeting functionality and facilitate receptor-mediated cellular uptake.

Compared with PLLA-based materials, PDLLA is amorphous because it contains both D- and L-lactide units, which can provide different degradation and drug-release characteristics.
