Description

General Description

ALC-0315 is an ionizable amino lipid specifically designed for nucleic acid delivery applications. During LNP formulation at acidic pH, ALC-0315 becomes positively charged, facilitating efficient complexation and encapsulation of negatively charged mRNA molecules. Under physiological conditions, the lipid remains largely neutral, minimizing toxicity and improving biocompatibility.

Following cellular uptake, the acidic endosomal environment protonates the ionizable amine groups, promoting endosomal membrane disruption and efficient release of mRNA into the cytoplasm. This mechanism contributes significantly to the high transfection efficiency observed in mRNA-LNP systems.

ALC-0315 has become one of the most important ionizable lipids in modern mRNA therapeutics and serves as a benchmark lipid for the development of next-generation lipid nanoparticle delivery systems.
