General Description

PLLA-PEG-Vinyl Sulfone (PLLA-PEG-VS) is a heterobifunctional biodegradable block copolymer consisting of poly(L-lactide) (PLLA), a hydrophilic polyethylene glycol (PEG) spacer, and a terminal vinyl sulfone (VS) functional group.

The hydrophobic PLLA block provides a biodegradable polymer domain suitable for forming polymeric nanoparticles, micelles, and other self-assembled nanostructures. The PEG segment improves aqueous dispersibility and colloidal stability while presenting the reactive vinyl sulfone group at the surface of the resulting nanocarrier.

Vinyl sulfone is an electrophilic functional group that reacts efficiently with thiols (–SH) through Michael-type addition to form stable thioether bonds. This chemistry is particularly useful for conjugating cysteine-containing peptides, proteins, antibodies, and other thiolated molecules.

Compared with some other thiol-reactive functionalities, vinyl sulfone offers good stability in aqueous environments while retaining efficient thiol reactivity.
