General Description
Fmoc-NH-PEG-OH is a versatile heterobifunctional PEG building block designed for orthogonal synthetic strategies and solid-phase peptide synthesis (SPPS). The terminal Fmoc-protected amine remains stable under acidic conditions and can be selectively deprotected under mild basic conditions, typically using 20% piperidine in DMF, to generate a free primary amine (NH2-PEG-OH) for subsequent conjugation.
The terminal hydroxyl group serves as a convenient synthetic handle and can be converted into a wide variety of functional groups, including carboxyl, NHS ester, maleimide, azide, alkyne, aldehyde, acrylate, methacrylate, epoxide, and thiol.
