General Description
NH2-PEG-COOH (Amine-PEG-Carboxyl) is a versatile heterobifunctional PEG linker designed for orthogonal bioconjugation and polymer functionalization. The terminal primary amine reacts efficiently with NHS esters, activated carboxylic acids, aldehydes, isothiocyanates, epoxides, and other electrophilic functional groups, allowing selective conjugation to proteins, peptides, polymers, nanoparticles, and biomaterial surfaces.
The terminal carboxylic acid group can be activated using carbodiimide chemistry (EDC/NHS) or peptide coupling reagents such as HATU or HBTU for reaction with primary amines, providing an additional orthogonal conjugation site. The presence of both amino and carboxyl functionalities makes NH2-PEG-COOH an excellent intermediate for the synthesis of multifunctional PEG derivatives.
The hydrophilic PEG spacer enhances water solubility, molecular flexibility, and biocompatibility while minimizing steric hindrance and reducing nonspecific protein adsorption.
