General Description
PLLA-PEG-Maleimide (PLLA-PEG-MAL) is a heterobifunctional biodegradable block copolymer consisting of poly(L-lactide) (PLLA), a hydrophilic polyethylene glycol (PEG) spacer, and a terminal maleimide (MAL) functional group.
The hydrophobic PLLA block provides a biodegradable polymer domain suitable for forming nanoparticles, polymeric micelles, and other self-assembled nanostructures. The PEG segment improves aqueous dispersibility and colloidal stability while presenting the maleimide functionality at the surface of the resulting nanocarrier.
The terminal maleimide reacts selectively with thiol (–SH) groups, particularly cysteine residues in peptides and proteins, under mild conditions to form stable thioether linkages. This chemistry provides a convenient approach for attaching antibodies, peptides, proteins, targeting ligands, and other thiol-containing molecules to PLLA-PEG-based nanoparticles and biomaterials.
PLLA-PEG-MAL is particularly useful for preparing ligand-targeted biodegradable nanoparticles, functional polymeric micelles, polymer-bioconjugates, and advanced drug delivery systems.
