Description

General Description

DMG-PEG-Mannose is a functional lipid-PEG conjugate consisting of a dimyristoyl glycerol (DMG) lipid anchor, a hydrophilic polyethylene glycol (PEG) spacer, and a terminal mannose ligand.

The hydrophobic DMG moiety enables incorporation of DMG-PEG-Mannose into lipid nanoparticles, liposomes, micelles, and other lipid-based formulations. The PEG chain provides a hydrophilic spacer that presents the terminal mannose group at the nanoparticle surface.

Mannose is widely investigated as a targeting ligand for mannose-binding receptors, including the mannose receptor CD206, which is expressed on several immune-cell populations such as macrophages and dendritic cells. Surface presentation of mannose can therefore be used to investigate receptor-mediated cellular uptake and targeted delivery.

Compared with phospholipid anchors such as DSPE, the DMG lipid anchor has shorter C14 hydrocarbon chains and can exhibit more dynamic association with lipid nanoparticle membranes. This property makes DMG-PEG derivatives particularly useful in the design and optimization of PEG-lipid-containing LNP formulations.
