General Description
HO-PEG-COOtBu is a versatile heterobifunctional PEG building block designed for orthogonal synthetic strategies. The terminal tert-butyl ester-protected carboxyl group remains stable under neutral and basic conditions but can be readily deprotected under acidic conditions (typically trifluoroacetic acid or HCl in dioxane) to yield HO-PEG-COOH.
The terminal hydroxyl group serves as a convenient synthetic handle and can be readily converted into a variety of functional groups, including amine, NHS ester, maleimide, azide, alkyne, aldehyde, acrylate, methacrylate, epoxide, and thiol. This enables the sequential preparation of a broad range of customized PEG derivatives.
The hydrophilic PEG spacer improves water solubility, molecular flexibility, and biocompatibility while reducing steric hindrance and minimizing nonspecific interactions.
