General Description
mPEG-OPSS is a highly versatile monofunctional PEG reagent designed for reversible thiol-selective bioconjugation. The terminal methoxy group is chemically inert, preventing polymer crosslinking, while the ortho-pyridyl disulfide (OPSS) group serves as a highly selective thiol-reactive functionality.
The OPSS group reacts rapidly with free sulfhydryl groups on cysteine-containing proteins, peptides, antibodies, enzymes, and thiolated nanoparticles through thiol-disulfide exchange, forming cleavable disulfide bonds while releasing pyridine-2-thione. These disulfide linkages are stable extracellularly but can be cleaved in intracellular reducing environments, making mPEG-OPSS particularly valuable for redox-responsive drug delivery and controlled release systems.
The PEG spacer enhances aqueous solubility, improves biocompatibility, reduces nonspecific protein adsorption, and prolongs circulation time in vivo. mPEG-OPSS is widely employed in protein PEGylation, antibody conjugation, nanoparticle surface engineering, reversible protein immobilization, biosensors, and advanced biomedical research.
