General Description
OPSS-PEG-COOH is a versatile heterobifunctional PEG linker designed for orthogonal bioconjugation and reversible thiol coupling. The terminal OPSS (ortho-pyridyl disulfide) group reacts rapidly and selectively with sulfhydryl (thiol) groups on cysteine-containing proteins, peptides, antibodies, enzymes, and other biomolecules through thiol-disulfide exchange, producing stable disulfide linkages while releasing pyridine-2-thione as the reaction by-product.
The terminal carboxylic acid group can be readily activated using carbodiimide chemistry (EDC/NHS) or peptide coupling reagents to react with primary amines, allowing efficient conjugation to proteins, peptides, polymers, nanoparticles, and biomaterial surfaces.
The hydrophilic PEG spacer enhances water solubility, molecular flexibility, and biocompatibility while reducing steric hindrance and minimizing nonspecific protein adsorption. OPSS-PEG-COOH is extensively employed in reversible protein conjugation, antibody modification, hydrogel synthesis, nanoparticle engineering, targeted drug delivery, and stimuli-responsive biomaterials.
