General Description

PCL-PEG-OPSS is a biodegradable amphiphilic block copolymer developed for advanced drug delivery and thiol-specific bioconjugation. The hydrophobic poly(ε-caprolactone) (PCL) block readily self-assembles into polymeric micelles and nanoparticles capable of encapsulating hydrophobic therapeutics, while the hydrophilic polyethylene glycol (PEG) block improves water solubility, colloidal stability, prolonged circulation, and biocompatibility.

The terminal ortho-pyridyl disulfide (OPSS) group undergoes rapid and selective disulfide exchange with free sulfhydryl groups on peptides, proteins, antibodies, cysteine-containing ligands, and other thiolated biomolecules. This reaction forms reversible disulfide bonds while releasing pyridine-2-thione, allowing efficient conjugation under mild conditions.

Because the resulting disulfide linkage is cleavable under intracellular reducing conditions, PCL-PEG-OPSS is widely used for preparing redox-responsive nanoparticles, targeted drug delivery systems, antibody-polymer conjugates, peptide-functionalized nanocarriers, and other advanced biomaterials.
