General Description
mPEG-Succinimidyl Succinate (mPEG-SS) is a highly reactive linear monofunctional PEG reagent designed for efficient amine-selective PEGylation. The terminal methoxy group is chemically inert, preventing polymer crosslinking, while the succinimidyl succinate group serves as an activated NHS ester for rapid conjugation with primary amines.
The activated succinimidyl succinate reacts readily with proteins, peptides, antibodies, enzymes, amino-containing polymers, nanoparticles, and other biomolecules under mild aqueous conditions (typically pH 7.2–8.5), producing stable amide bonds. The succinate spacer provides flexibility between the PEG chain and the conjugated molecule, helping reduce steric hindrance and improve conjugation efficiency.
The PEG spacer enhances aqueous solubility, improves biocompatibility, reduces immunogenicity, minimizes nonspecific protein adsorption, and prolongs circulation time in vivo. mPEG-SS is widely employed for therapeutic protein PEGylation, enzyme modification, antibody conjugation, polymer synthesis, nanoparticle surface engineering, and advanced biomedical research.
